Methods

-A comprehensive measure of prenatal maternal stress was con- structed based on an existing measure developed in ALSPAC (Cecil et al., 2014). The prenatal stress measure included 55 maternal-reported items collected through questionnaires administered between 8 and 32 weeks of pregnancy.

-Serum concentrations of IL-6 and CRP were examined in children at 9 years of age (mean ± SD = 9.85 ± 0.31 years). Blood samples were drawn from non-fasting participants, centrifuged, and frozen at − 80 ◦C in 1 mL aliquots. IL-6 and CRP concentrations were assayed in 2008 after a median storage period of 7.5 years.

 

The researchers created a test to measure the stress which included questions for the mothers to answer during 8-32 weeks. The test was used to see how much stress the mother was feeling as well as what type of stress. They used this from another study, Alspac.

The researchers tested the children’s blood at age 9 to measure IL-6 and CRP, which are indicators of inflammation in the body. After collecting the blood, they separated and stored it at very low temperatures until it was analyzed. Measuring these biological markers allowed the researchers to examine whether prenatal stress could have long-term effects on a child’s immune functioning.

Introduction

-Several studies have demonstrated associations of prenatal maternal stress including stressful life event and maternal psychopathology, including stressful life events and maternal psychopathology, with children’s mental health from early childhood into adulthood including depression and externalizing behaviours.

-Prenatal stress exposure can affect children’s endocrine and immune system function, which an explain the finding in some studies that c children whose mothers experiences stress of adversity during pregnancy have higher concertation’s of immune biomarkers.

 

Research has shown that when mothers experience high levels of stress during pregnancy, such as major life events or mental health problems, it can affect their child’s mental health. These effects can last from early childhood into adulthood and are linked to issues like depression and behavioural problems such as acting out.

Stress during pregnancy can affect how a child’s hormone and immune systems develop. This may explain why some studies have found that children whose mothers experienced stress while pregnant have higher levels of inflammation markers in their bodies.

 

 

Results

Prenatal maternal stress, child IL-6 and CRP concentrations, and youth-reported symptoms of GAD and depression. Prior to and after adjustment, higher prenatal maternal stress was strongly associated with more symptoms of GAD (adjusted b = 5.75, SE = 2.02, p =.005) and depression in adolescence (adjusted b = 19.12, SE = 3.69, p <.001), and with higher concentrations of IL-6 in childhood (adjusted b = 1.80, SE =.65, p =.006). Higher concentrations of IL-6 in childhood were associated with increased symptoms of depression in adolescence.

 

The researchers looked at stress during pregnancy, levels of inflammation in children, and symptoms of anxiety and depression in adolescence. They found that when mothers experienced higher stress while pregnant, their children were more likely to report symptoms of anxiety and depression as teenagers. Higher prenatal stress was also linked to higher levels of IL-6, which is a marker of inflammation, at age 9. They also found that kids with higher IL-6 levels were more likely to have depressive symptoms later on.

Discussion

Although no mediation of the associations between prenatal maternal stress and children’s depression or GAD through immune marker concentrations was observed, this work adds to the emerging evidence base examining associations between prenatal maternal stress with children’s inflammation and mental health.

Even though the researchers did not find that inflammation directly explained the connection between prenatal stress and later anxiety or depression, the study still adds to existing research. It supports the idea that stress during pregnancy is linked to changes in children’s inflammation and mental health, even if the exact pathway is not fully clear.

The inconsistent results across studies may be because researchers measure prenatal stress in different ways. In this study, the researchers used a broad and detailed measure of stress that included life events, depression, substance use, financial problems, and relationship issues, which may explain why their findings differed from some other studies.

 

Future Directions

It would have to have a different technology to measure how badly the stress is affecting the child during pregnancy.

    o I believe that future research should follow up with the children in adulthood to understand how the inflammation connects to the children’s outcomes in life. It would be interesting to see if children who had higher inflammation levels received less education or had lower income throughout adulthood. It would also be interesting to do a retrospective study to understand the specific type of stress the mother was under during her pregnancy (E.g., financial, substance use, stressful life events). This may be used to understand how different types of stress affect the inflammation levels in the children.

    o One concept that I found difficult was understanding IL-6 and what the levels meant for the study. IL-6 or interleukin-6 is a protein made by the body’s immune system. It is used as a biological marker, as the levels of IL-6 increase when the body is in an inflamed state. An inflamed state is the body’s natural response to injury, infection, and stress, as a part of the body’s immune defence system. IL-6 acts like a messenger, indicating to other immune cells that something is wrong and that more immune cells need to be made to fix the problem. During stress, the body is on high alert, preparing for something to be wrong, even if the source of the stress isn’t internal. The body is preparing for some kind of threat, hence the higher levels of IL-6. This means that the children whose mothers were more stressed during pregnancy had higher IL-6, which made their bodies more primed for inflammation, which could also make them more vulnerable to mental health issues later. experiencing higher levels of stress, and their bodies are constantly on high alert in life.
